The parish was formed by Order in Council 27 Aug 1846 (London Gazette, 22 Sep 1846)
Premises in City Road and River St. were used before the foundation stone of a permanent church was laid on 16 Jun 1851, and the building consecrated on 20 May 1852. (see Archdeacon Rushton's Visitation Returns, 1846 MSf 942.72 R121 Vol. 45) It was damaged by the air raids of Christmas 1940 and pulled down in 1948.
By Order in Council, 8 Dec 1950 (London Gazette, 12 Dec 1950) St. Mark's was united with St. Steven's, Hulme, as the parish of St. Stephen's with St. Mark's, St. Steven's becoming the parish church.
